Initial OD = 0.775 and the dilution is 1:10
By diluting in 1:10 the original sample will get 10 times low OD than the original without diluting. So, the original sample has the OD 0.775 x 10 = 7.75

O1 V1 = O2 V2 formula is use here, where O is using for Optical density and V for volume.
O1 = 7.75, V1 = ‘X’ ml, O2 = 0.1, and V2 = 40 ml
7.75 x X = 0.1 x 40
