How could one separate one type of nucleotide, ie: dgtp, datp, catp from a pellet of nucleic acids extracted from yeast?

Solution

Nucleic acids are soluble in water, because of these charges, polar molecules like DNA or RNA can interact electrostatically with water molecules allowing them to dissolve in water.

The nucleotides in the cell pellet can be separated by adsorption onto dowex-1 ion exchange resin column followed by elution with dilute hydrochloric acid and lithium chloride.

After the nucleotides are separated, lithium chloride was removed by extraction with organic solvents. The overall yield of nucleotides was 75-80%.
