You have a microtube containing 1 mL of a solution with 4.3 x 104 cells/mL and you are to produce a solution that contains 43 cells/mL. What serial dilutions must you perform?

Solution

Initial cells = 43×103

Using serial dilution method:

Ist dilution: (100+900 ) microliters = 1000 microliters = 1 ml.

So it will be final /sample = 1000/100= 10 = 1/10 dilution.

So cells will be 43 ×102 in this sample.

in te next dilution, same process is repeated. And the dilution will be then 1/100 so cells will be 430

and finally in 1/1000 dilution the cell count will be 43 cells/mL.
