Calculate the estimated concentration and yield of a 1 mL DN

Calculate the estimated concentration and yield of a 1 mL DNA sample, if absorbance at 260nm is 0.862 when you dilute it 1:200 and test it using the spectrophotometer.

Formulae for calculation: Concentration (ng/HL) Abs@260 x 50 x dilution factor DNA yield (ug) DNA concentration x total sample volume (ml)

Solution

according to your question the answer will be this one

Ans is -DNA yield in(g) = 0.2115 × 10–3

first we need to find out concentration in (ng/l) so our abs at 260 is 0.862 multiplied with 50 × dilution factor is given(1:200), after the calculation the concentration of dna is 0.2155 × 109

than we need to find out DNA yield in g, so 0.2155 × 109 is divided by 106 for unit conversion

and finally we get DNA yield in g is 0.2115 × 10–3g
