In order to prepare 20L of 1X TAE buffer from 50X TAE stock
In order to prepare 20L of 1X TAE buffer from 50X TAE stock solution, how much (in ml) of the 50X TAE buffer is diluted with water to prepare 20L of 1X TAE?
In order to prepare 20L of 1X TAE buffer from 50X TAE stock solution, how much (in ml) of the 50X TAE buffer is diluted with water to prepare 20L of 1X TAE?
Solution
Llet us see the logic of this.
50x means 50 times concentrated, and 1x means 1 time concentrated. The solutions what we use regularly are all 1X. higher X is made to store as stock for future use. So if you dilute 50x to 1x we have to add, 50 times water is added. if you take 1 L of 50x TAE and dilute it upto 50L, then it becomes 1X solutions.
Now let\'s solve this using formula.
This can be solved by formula RT / G .
Required concentration = 1 x =R
Given concentration = 50x =G
Total volume = 20L =T
SO, RT/G
= (1)(20L) / 50
= 0.4 L
total is 20 L , in that 0.4 L should be TAE, so remaining 19.6 L is water.
