What are some solutions for recovering information from degraded DNA? Why are more polymorphic loci useful in identifying a mixture? What are some of the difficulties with low copy number DNA analysis? What kinds of checks and balances can a laboratory employ to ensure reliability of DNA profiles coming from low amounts of DNA profiles coming from low amounts of DNA template?
Solution
1- The probability of recovering information from a degraded DNA is very less, but you can give a try by diluting the PCR up to 1:10 or more and you can use that diluted product as template. This method is applied when you have a faint bands.
2- By polymorphic loci the PCR product will be very small and markers can easily work with more degraded DNA samples.
They can be more easily available in genome.
There will be less or no stutter products.
And also the sampling processing may be done completely automated.
3-The normal difficulties faced during low copy DNA analysis are
Stuttering will happen which inturn causes the problem of extra magnification, because this will lead to increase in confusion between stutters and DNA profile.
Since we have low number of copies we have to take care to reduce contamination.High care should be taken in all stages of collection and processing.
And also we have problem of lack of research in low copy DNA analysis because of that we lack techniques in this type of circumstances.
4-The best laboratory techniques employed to check the reliability of DNA profiles coming from low amount of DNA profiles and DNA templates are
Have to construct calibration curves at low DNA concentration by statistical methods.
Before getting in to any detection we have to make sure about detection probability and detection limits or else our samples may get wasted.
And also as said earlier we should be highly alert in order to avoid contamination of sample as they are highly sensitive.
