Predict the effect of the NADPH/NADP+ ratio (as compared to controls) in cells under the following circumstances:
a. Withdrawal of serum.
b. Addition of DHEA.
c. Addition of H2O2.
d. Withdrawal of serum and addition of H2O2
Solution
a. Withdrawal of serum : NADPH/NADP+ ratio decreases as glucose-6-phosphate dehydrogenase (G6PD) enzyme activity decreases with withdrawal of serum.
b. Addition of DHEA :There is a decrease in NADPH/NADP+ ratio as DHEA inhibits glucose-6-phosphate dehydrogenase (G6PD).
c. Addition of H2O2.NADPH/NADP+ ratio increases as addition of H2O2 enhances the activity of pentose phosphate pathway thereby increasing the NADPH/NADP ratio.
d. Withdrawal of serum and addition of H2O2: Not much change in NADPH/NADP+ ratio.
