(2) the substrate and the enzymes should be able to undergo the needful confitmational changes for for proper binding..

(3) the binding should be no-covalent in nature so that the substrate should easily be removed after the process done to allow the enzymes to bind the other substrate..

(4) the Km value of the enzymes should be less to allow the binding at a lower substrate concentration too..
