DNA synthesis takes place in 5’-->3’ direction. DNA Polymerase forms two covalent phosphodiester bonds between 3\' hydroxyl ends of one nucleotide, (\"acceptor\") with the 5\' phosphate end of another (\"donor\"). For the same DNA polymerase require free 3’-OH group.
In the absence of 3’-OH group phosphodiester bond formation is not possible and DNA synthesis stops.
