Answer the following questions and attach with your pre-lab write-up. Assume that you have collected 35 ml egg white. Answer the following questions: a) How much 1.0 N Acetic acid will be added to the above egg white? What is the purpose of adding acetic acid? Explain briefly. b) What will be the pH of the egg white solution after adding I N acetic acid. c) Which salt would you use for this experiment? Why this salt is useful compared to other salts? d) Which three major proteins are found in egg white? Out of these three proteins, which protein requires lower ionic strength and higher ionic strength to precipitate out? e) Convert 10,000 RPM into RCF. Given that the radius of rotor is 10.5 cm.
Solution
(a)2 drops of 1.0N acetic acid should be added. This can be explained by the reaction between acid and egg white which causes protein denaturation by altering the electrostatic interactions. The main effect is unfolding the tertiary conformation of peptide chain, decreasing of solvation and subsequent decrease of solubility.
(b)7.6 TO 9.7
(c)Details of experiment and purpose not given.
(d)The major proteins in egg white are ovalbumin, conalbumin, ovomucoid, and lysozyme.
(e)RCF=1.12 x Radius x (rpm/1000)2 ; So here,RCF=11760
