You want to determine what the optimal pH is for E.coli to grow. How would you set up the experiment? Give a detailed explanation!

To determine the optimal pH , experiment will be set up in different pH conditions. Growth medium for E. coli will be prepqred in a range of pH like 5.5, 6,6.5, 7, 7.8,,8 etc.and then the culture will be sreaked or inoculates in the media and left for growth at same temperature and same time duration.

If one culture is grown overnight at 37degres celcius, then all culture would be given same incubation conditions.

Then the growth in each pH condition will beobserved and whichever culture will show best grpwth, that will be optimal .

If two different cultures for example at pH 6 and 6.5 show almost equal growth then that experiment will be repeated for a range between these two pH values say , 6.1,6.2,6.3,6.4,6.5

Again the culture that exhibits best growth, will be selected as optimal pH for its growth.
