Taq DNA polymerrase is an enyzme isolated from the thermostable bacteria Thermus aquaticus. Tad Pol is very much important in PCR because of its thermostability. its optimum temparature range is 75–80 °C and has a half life of 92.5 °C for 2 hours. In a PCR sample, DNA fragments of E. coli will be present and this may cause a false amplification. to remove this DNA fragment DNAse enzyme can be used. But if this DNAse is not removed, it could damage the primer. Therefore heating the sample up to 75°C will remove all the protein contaminants except our Taq DNA polymerase because of its thermostability. Centrifugation, filtration, and dialysis can remove all the presipitated contaminanys later.
