The pure culture is derived from mixed culture  by transferring a small sample into a new sterile growth medium in a manner that disperses the individual cells across the medium\'s surface. for the purpose of making the pure culture medium agarous gel is used for gelling pure culture can be grown in petri dishes or test tubes by thinning the sample by innoculating them in the new medium.
