1.) Name the top layer of the hematocrit? Indicate what component(s) is/are absent from this top layer that are present in whole blood?
2.)Can you describe a circumstance when a patient may need blood plasma and not blood serum?
Solution
1. Hematocrit is a diagnostic method for the analysis of the blood. The blood which is in a calibrated tube, allowed to stand for one hour, the sedimentation rate of blood cells (settling out of blood cells from plasma) is determined after the centrifugation of tube. The blood contents are separated in to three layer, upper layer contain plasma, middle layer contain WBCs and platelets and bottom layer contain packed RBCs.
Upper layer of hematocrit doesn\'t contain any blood cells like RBC, WBC or Platelets.
Plasma = Blood - Blood cells. The composition of plasma layer is water, dissolved blood proteins like serum albumin, globulin and fibrinogen, clotting factors, electrolytes (Na+, K+, Cl-, Ca2+ etc.), and hormones,
2. Serum is similar in composition of plasma but it doesn\'t contain any clotting factors. generally it is used for checking of the blood groups and diagnosis of the disease. Whereas plasma is transfused in to patient who lack the blood cells. In hemophilia B disease condition, it is necessary to deliver the fresh frozen plasma to the patient.
