Im working with plasmids PGL4.75 and PGLO. My proffesor asked me to mimic the expression of PGLO in eukaryotic cells, as it was performed in prokaryotic cells (hands on and online) , but im not sure if she means the procedure or something else

PGLO is widely used to develop genetically modified organisms. PGL4.75 contains luciferase reporter gene. This plasmid is used for high expression and reduced anomalous transcription. The plasmid has a codon optimized synthetic gene and consensus regulatory sequences. So PGL4.75 is highly useful for expressing in eukaryotic cell specially mammals. The professor had meant to say to develop PGLO in such a way that it can be expressed in eukaryotic cell as it express in prokaryotic. PGLO contains GFP, bla gene, araC, pBAD promoter and multiple cloning sites. These help in replication of the plasmid DNA and GFP expression in bacteria subsequent transformation. Mimicking the expression as prokaryotic cell means performance of plasmid would same in eukaryotic cell too.
