To isolate Neurosporene (haploid) auxotroph\'s from prototroph, you perform a large-scale mutagenesis. You learned in class that you should culture them on the complete medium first and then test them on the minimal media plate by making replica plates. But, there is a way to do it using the minimal media plate ONLY. Dr. T laid out a basic plan below. He left out culture conditions of Neurosporene for this experiment. Minimal Medium contains the glucose which is main source of carbon. Metabolism of glucose can generate the ATP. Ammonium is main source of nitrogen, from their cell can synthesize required amino acid.

Mutation in microorganism culture alters the key feature which helps in survival. Cells will unable to synthesize necessary metabolites after mutation. But if Media containing required components then mutated microorganism can use those components from media.

In replica plate technique multiple colonies grow in rich in nutrients. After that a paper will touch on surface of media. And Re-touch the used side of paper on the new media plate. Cell will grow if they will able to synthesize required component or component is available in media. Mutant microorganism will not able to grow in deficient media. This method used to isolate for different bacterial nutritional auxotroph.
