You are performing a PCR to determine if your gene of interest has been successfully incorporated into a plasmid. Is a high fidelity polymerase necessary? Why or why not?
Solution
The fidelity of polymerase enzyme is very important in PCR, why because it had several steps involved to produce the exact sequence of desired template. DNA polymerase should add correct nucleotides, should read the template strand, and should select the appropriate nucloside triphosphate. More than that they should perform the proof reading to eliminate the wrongly placed nucleotides. Therefore the fidelity of polymerase is very important in PCR
