The article mentions about the white nose disease caused due to the growth of fungus Pseudogymnoascus destructans on bats. This fungus and cells of bat gets attacked and infected by a virus called as gammapartitivirus which contains ds RNA (NOMENCLATURE not so important FOR VIRUS STRAIN AND FUNGUS).
Further, transmission electron microscopy was done to confirm the infection by virus and fungus. The sample has mixed strains of virus and fungus.
Other experiments were performed on fungus strain and virus strain for example---
1)RT-PCR was done for the RNA isolated from fungus samples for genes like glyceraldehyde-3-phosphate dehydrogenase, elogation factor- alpha, internal transcribed spacer (ITS) with random primers.
2)the terminal ends of RNA of virus mRNA were sequenced using RNA ligase and linker sequences of nucleotides complementary to the ends of mRNA.
3)fungus strain was grown on SB-agar media and as agar is a polymer of agarose-agaropectin which is complex network of carbohydrates which has a property to retain water called as water potential calculated by using eletrodes with buffer salts in agar as compared to polyethylene glycol water absorption levels.
4)bioinformatics software analysis was done to study phylogenetic linkage of the fungus and it revealed its similarity with penicillium fungus and virus that infects penicillium fungus.
5)the hyphae of fungus and conidia were grown in proper media and after the growth from fungal spores, membrane transfer was done to transfer the fungus hyphae to new SB-agar plates.
6)Further ,centrifugation was done to isolate virus strain in sodium phosphate buffer.
7)the fungus was cultured by protoplast separation method, hyphael culture method and studied under electron microscope to study the infection with virus particles.
8)heat treatment was given to fungal hyphae and further studies were done in this strain as compared to original strain.
9)the growth of virus was inhibited by using ribovirin, and cycloheximide.
10)INFERENCE-this strain of fungus and virus mixed infection leads to the killing of bats in north america and the disease is called as white-nose disease.
