Make 1L of 1x PBS. Final concentration of reagents are 150mM NaCl, 20mM KCl, 11.5mM Na2HPO4, 8.5mM NAH2PO4.

Solution

In first step, we dissolve the following ingredient into the 800 ml of distilled water.

Then, adjust the pH of solution at 7.4

Adjust volume upto 1L with additional distilled water

Sterlize the solution by autoclaving

For preparation of 1X Phosphate buffer saline, we calculate the amount of ingredient we are required to add into the water.

NaCl has molarity 150mM

Molarity (M)= moles of the solute per liter of solution

Moles of solute given by mass/ molecular weight (mw)

Here the formula is M = mass/mw L

Volume of solution is 1L

150 x 10-3 mol = mass/ 58.44x 1

= 8.7 gm

KCl = mw =74.55 gm/ml

Na2HPO4 = 141.96 gm/ ml

NaH2PO4 = 119.98 gm/ml

Using the same formula, concentration of ingredient is obtained that are given below

NaCl = 8.7 gm

Kcl = 1.5 gm

Na2HPO4 = 1.704 gm

NaH2PO4 = 1.08 gm
