What absorbance would you expect if the concentration of a p
What absorbance would you expect if the concentration of a protein was 5 M, and it has 8 tryptophans and 10 tyrosines?
Solution
to calculate absorbance we required molar extinction co-efficient
Molar extinction co-efficient = (No. of tryptophan residues * 5500) + (No. of tyrosine residue * 1490)
so accroding to that molar extinction co -efficient = (8*5500) + (10* 1490) =44000+14900=58900.
Molar extinction coefficient is the absobance of the 1 molar solution of the protin. In the above problem the conccentation of protine solution is 5 M it means it eqals to 5000000 M.
If the wavelength is 280 nm in the problem it is not specified so if we assume that wavelength is 280 nm and the molecular weight of trypto[phan and tyrosin is accroding to standard than above problem can be solvved. but it is not specified so furthe calculation can not be possible using assumption.
