Explain why the following steps are essential during sub- culturing: Flaming the inoculating instrument prior to and after each inoculation. Holding the test tube caps in the hand during transferring. Cooling the inoculating instrument prior to obtaining the inoculum. Flaming the neck of the tubes immediately after uncapping and before recapping.

1) This step flaming the inoculating instrument prior to and after each inoculation plays an important role because it mamaintains the sterility.

2) we hold the test tube caps in the hand with proper gesture of fingers and carefully because there are bacteria and contamination on our hand and won\'t want to contaminate the caps and tubes.

3) If we won\'t cool the incinoculating instrument prior to obtain the inoculum, excess heat can fixate the sample or kill the organorganisms.

4) If there are microbes on the neck of the tube , you don\'t want to accidentally bring those into the nutrients base , also you don\'t want to spread the microbes you were incubating by spreading it through the neck.
