A hanging drop slide slide is prepared to visualize the motility of bacteria microscopically. In this method, a ring of petroleum jelly is made carefully on a flat or depressed slide. A drop of living culture is then placed on it and a coverslip is placed above it. The slide is reversed and placed under a microscope for visualization of living microbes.
Most of the bacteria are visible under a microscope using this system but sometimes, some bacteria become poorly visible. In order to improve visualization, an oil-immersion system can be introduced in the microscope. This will improve visualization by preventing un-necessary scattering of light from the sample and thus most of the bacteria would be visible. Along with it, the brighness of the lamp may also be adjusted along with oil-immersion system to further enhance clarity of vision.
