Double stranded DNA molecules are amplified through polymerase chain reaction (PCR). In this technique, the two strands of DNA are unwound by heating to high temperature of 900C. Then, primers are designed to complementary to 3` end of sequnces and annealed. The primers are extended by DNA polymerase enzyme producing 2 different double stranded DNA molecules. In this, one cycle produced 2 DNA molecules, two cycles produce 4 DNA copies and 3 cycles produce 8 DNA copies and so on. Therefore, the number of cycles required to obtain double stranded amplification product is 1.
