You have generated a T cell clone in which LAT is knocked out (missing). you stimulate that cell with an epitope loading not the appropriate MHC and test for the IL-2 protein production. Do you expect to detect IL-2 or not? Explain

Solution

Linker for Activation of T cells (LAT) gene plays an important role in the downstream signalling of T-CELL receptor after T-CELL receptor recognises antigen by antigen presenting MHC 1 complex since even if we stimulate it with epitope directly, without MHC CLASS 1. We wont be getting the release of INTERLUKIN 2 because costimulatory signals between antigen presenting cell and T-cell would be missing. AND WITHOUT THOSE CO-STIMULATORY SIGNALS EVEN IF ANTIGEN IS PRESENTED TO T-Cell RECEPTOR DIRECTLY IT WONT GET ACTIVATED AND EVEN IF RECEPTOR GETS ACTIVATED SOME HOW, DOWNSTREAM SIGNALLING WONT OCCUR DUE TO MISSING LAT GENE

Hope that answers the question.. Feel free to comment and Discuss .... I shall be glad to help you in your related querries.
