An important trait that some bacteria have is resistance to tetracycline (a broad-spectrum antibiotic). Other bacterial strains are inhibited by tetracycline (they cannot reproduce and form colonies in its presence). Your challenge: diagram and briefly describe an experiment to test the hypothesis that the tetracycline resistance is a trait that is inherited and encoded by DNA. Hints: look to the experiments of Griffith and Avery, MacLeod, and McCarty for guidance; ideas for materials: Tetracycline-resistant bacteria culture (Tcr cells), tetracycline-sensitive bacteria culture (Tcs cells), agar plates with tetracycline, test tubes, boiling water.

Solution

Step 1: you need to prepare 4 agar plates containing media with presence of tetracycline.

step 2: Grow the two bacterial culture(Tcr and Tcs cells) in test tube over night in 37 degree Centrigrate. next daymake two set.

Set 1: Take a loop of overnight culture and strick the both culture Tcr cells and Tcs cells into two individual agar plates with proper marking. keep the both plates at 37 degree Centrigrate for overnight.

Set 2: Boil the both cultures and make two strick plates same as set 1.

step 3: next day you will find colonies in Tcr cells because it is Tetracycline resistant . but you will get no colonies at Tcs plate in set 1 experiments.

In set 2 experiments there will be no colonies in both plates, because due to boil, the genetic meterials(DNA) of the cell are disruped these is called Heat killed bacteria. so we can say that tetracycline resistance is a trait that is inherited and encoded by DNA.
