Poly(DL-lactic-co-glycolic acid)/poly(ethylene glycol) blends are being studied for manufacture of biodegradable micro particles to be used for controlled parenteral delivery of IgG antibody. The first step in preparing the microparticles is to make up a mixture containing .2% antibody, 18% polymer blend, 9% water, and 72.8% dichloromethane. Available for this procedure is 1 kg of a blend of 1% polyethylene glycol (PEG) in poly(DL-lactic-co-glycolic acid) (PLGA), 4kg of a solution of 15% PEG in dichloromethane, and a stock of pure dichloromethane. All of the PEG/PLGA blend and all of PEG/dichloromethane solution are used.

a) What quantity of pure dichloromethane is needed?

b) Antibody is added as an aqueous solution. What quantity and composition of antibody solution are required?

c) How much final mixture is produced?

d)What is the concentration of PEG in the final mixture?

Solution

a)- 3071.11gm

b)- quantity=88.88gm & composition= IgG + water

C)-8888.88

D)- 6.8%
