Considering the cultures used to inoculate each medium in this exercise, how many different microbial types should you expect to see on/in each medium? Explain your answer. Which medium was most difficult for you to transfer from? Which medium was most difficult for you to inoculate? Explain your difficulties. Did you notice a difference in density (turbidity) of growth in SB tubes inoculated from SB and NA slant? Suggest possible reasons why a difference might occur. Solution

Ans 1. Microbial growth can\'t be found from sterile NA/ NB .. If found then it has probable two cause a. The autoclaving was not done properly b. After autoclaving storage of plates is not done carefully(lead becomes open / lead wrongly tightened/ cotton plugging become loose etc).

Ans 2. Needs more information which types of culture you are using. And source of the culture.

Ans 3. Its your difficulty.. Although I had been facing problems future transfer from liquid to solid tube( making slant) , I can\'t balance two tube in one hand with the lead.. But now its gone with practice.
