Condensation often gathers in the bottom of agar slants. Why is it important in this exercise to limit condensation on plates but not on slants?
Solution
when pouring medium it should be at 40-45 degree centigrade. usually on plates condensed water will drop down on plate it will not allow to grow the colony evenly on the surface.
in case of slant there is slope so all water will gather near bottom that will not cause problem to grow colony at upper part.
it is better to maintain the temperature about 40-45 degree centigrade before pouring either on slant or petriplates to minimize water condensation.
and allow lid after solidification of medium, all process should be done in a good laminar flow cabinet.
