DCPIP is the only electron acceptor in the in-vitro experiment for light-dependent reaction of photosynthesis in chloroplasts. The electrons ejected by excited PSII and PSI and ultimately accepted by DCPIP, which gets reduces (so, gradually becomes colorless) as photosynthesis continues to occur.

In absence of DCPIP in the reaction tube, there is no other molecule to accept electrons ejected during photosynthesis. So, no electron is accepted in the tube.
