Where high fidelity PCR is required, why is it important to have the first few rounds of amplification be absolutely correct?

Solution

High fidelity PCR is done when after amplification or multiplication absolutely correct DNA sequences must be obtsined. This process is usually utilised during cloning analysis for studying protein expression of particular gene segments. In RT-PCR, RNA molecules are made and analyzed. Thus these RNA molecules need to be correct and obtained from high fidelity PCR. Several applications require multiplied DNA fragments where absolutely correct DNA segments are required and high fidelity PCR can be used for this purpose.

It is very important for the first few rounds of amplification be absolutely correct because if a few errors are introduced at the beginning , each error will be amplified exponentially along with the original DNA sequence during subsequent cycles. As a result if PCR with small amount of template DNA is started, any error during first few cycles would appear as a major error in final PCR products and would not serve the purpose of high fidelity PCR.
