Need help writing a one and a half page paper (250-300words) over the introduction of the hemoglobin electrophoresis assay. In this define what electrophoresis is, why is it important, what is it used for, the best methods it is used on, how this helps with certain diseases like sickle cell anemia, and how it is important in our time now. Solution

Haemoglobin is a red colour pigment present in the RBC of blood. It carries oxygen molecules efficiently while abnormal hemoglobin molecules cannot carry oxygen. hemoglobin F and hemoglobin A are the common types of haemoglobin found in blood. Abnormal types hemoglobin are S,C,D,E,M. Healthy adults usually have haemoglobin A when haemoglobin F level is more than 2% it indicates certain diseases

Definition: hemoglobin electrophoresis essay is used to measure different types of haemoglobin present in the blood based on the character of seperation at different electrical charges.

Importance: it is used to diagnose diseases in which abnormal hemoglobin is produced. Some of the important diseases diagnosed of Sickle Cell anaemia, thalassemia.

How it is done : first of all the blood is obtained from the patient with the help of a syringe and collected in a vial. The sample is then processed by machine. as mentioned above as different types of hemoglobin have different charges then move with different speeds in the gel. The process is done by using cellulose acetate at 150 to 200 volts. Finally the hemoglobins move on the gel and form bands .

It was found that high performance liquid chromatography HPLC will give good results for thid assay.

The mobility of the different types of hemoglobin is compared to those of a control sample the haemoglobin molecule is negatively charged and migrate towards anode.For the seperation of hemoglobins A C E S D citrate Agar gel electrophoresis at acidic pH is used.

Diagnosing diseases l:

It is done to find the types of haemoglobin present in the blood only used to diagnose anaemia such as thalassemia.

Haemoglobin S is abnormal form associated with the disease Sickle Cell anaemia, in which the blood blood cells have Crescent shape which can block small blood vessels. hemoglobinC is abnormal form associated with hemolytic anaemia. In normal healthy adults the percentages of hemoglobin will be Hb A- 95% Hb F- 1 to 2% HbS- 0% HbC-0% . In the infants and children the percentages are HBF in newborn 50 to 80% ,six months 8% ,HBF after 6 months 1 to 2%.

Sow hemoglobin electrophoresis is used to identify this is us like haemoglobin C diseaseS, rare hemoglobinopathy Sickle Cell anaemia, thalassemia.

Presence of hemoglobin S -Sickle Cell anaemia or thalassemia

2. presence of hemoglobin S and F Sickle beta Thalassemia (sickle hereditary persistence of fetal hemoglobin)

Uses:

Nowadays the hemoglobin electrophoresis is used to evaluate hemolytic anaemia ,anaemia related to iron deficiency , to check the family history of hemoglobinopathy, for positive neonatal screen results and positive results of Sickle cell history.

Automated HPLC instrument proves to be the best methods for hemoglobinopathy screening.
