1). treat the cancerous cell line with the appropiate concentration of the drug and take untreated cells as control

2).Isolate the total RNA of the treated and untreated cells with any RNA isolation kit.

3). to the Quantitative real time PCR to look for the mRNA level of cell cycle progression genes by using specific primers.

4). compare the mRNA expression of cell cycle progression genes with the untreated sample.

5). If the drug is working then mRNA expression of cell cycle progression genes will be less as compared to the control untreated cells mRNA.

6). You can also check the cell cycle progression of the treated cells by using flow cytometry and compare it with the untreated cells.
