Protein z can bind to two ligand named v and w. The two complexes have the following Kd values: Kd(zv) = 300 nM, and Kd(zw) = 0.3 mu M. Which ligand has the highest affinity for protein Z? Explain.

Solution

The smaller the KD value the greater the affinity of the antibody for its target. At equilibrium, the rate of [antibody] [antigen] complex formation is equal to the rate of dissociation into its components [antibody] + [antigen]. The measurement of the reaction rate constants can be used to define an equilibrium or affinity constant (1/KD).

So, reaction rate of V=1/Kd=1/300(300 nm=0.3 micrometer.)

reaction rate of W=1/Kd=1/0.3

so, both ligand have same affinity towards protein Z.
