what is the light adjustment for stained vs unstained specimens in a microscope
Solution
We use bright field microscopy tool in order to view the light or stained cells
In the bright field microscopy adjustmen we use the light from an incandescent source and this light is aimed toward a lens which is usually kept beneath the stage. This stage is called as the condenser. The next step is that The light then passes through the specimen, next it passed through an objective lens and finally it reaches to the eye through a second magnifying lens which is called as the ocular. This is for stained particles.
On the other hand we use dark field microscopy tool adjustment for unstained specimens .
In order to view something in dark field, we usually use a an opaque disc. Moreover, this disk is then placed underneath the condenser lens,This makes sure that only that part of light which gets scattered by the objects on the slide can reach the eye of the observer.. This means that instead of the light coming up through the specimen, it gets reflected by particles on the slide. This allows us to see unstained specimens.
