The steps are as follows:

pglo plasmid contains a gene for expression Green Fluoresecent Protein.The cells that will be transfected with pglo plasmid will express the GFP gene and GFP protein will be formed.GFP protein give a fluorescent green color. This will help to see the pglo expression.This can be done by FACS analysis which shows the percentage of GFP positive cells thus showing the transfection cells.
