Cell density = average of cells * dilution factor / volume of small square or middle square (ml)

Average =20+30+25+28/4=25.75

Consider your sample is not diluted then take dilution factor 1.

Volume of small square = width *height *depth =1*1*0.1mm³=0.0001ml

Cell density =25.75*1/0.0001 =257500 cells per ml

If you dilute your sample in the ratio 1:5 then dilution factor will be 5. So we have to multiply with 5 instead of 1.
