1. The concentration of gel determines the pore size of the gel which affects the migration of DNA. The resolution of the DNA changes with the percentage concentration of the gel. Increasing the concentration of a gel reduces the migration speed and improves separation of smaller DNA molecules, while lowering gel concentration permits large DNA molecules to be separated.

2. A concentrated gel is needed when the DNA to be separated has a very small size. If the concnetration of the gel is more that means the pore size is small, all the other impurities will not be able to pass through the gel and the DNA to be separated will be isolated.
