Electrophoresis has been helpful in separating subunits of proteins, nitrogen bases, DNA and RNA. Electrophoresis runs on the size of molecules and their polarity or charge. During electrophoresis, the molecule which has been loaded runs from top of \'-\' electrode of gel to bottom of \'+\' electrode of gel. The smaller molecules move faster and reach the bottom-line of \'+\' electrode. The larger molecules mostly lie on the top of gel near \'-\' electrode. PAGE or polyacrylamide gel electrophoresis uses commonly SDS as denaturing agent and this SDS given even negative charge to all molecules which have to be migrated within a gel. Bis-acrylamide and acrylamide together determine the pore size within a gel. If more the percentage of acrylamide, smaller will be the pores formed within a gel due to cross linking of acrylamide with bis-acrylamide. If the molecules which have to be migrated within a gel are smaller, then the percentage of acrylamide has to be higher. If the molecules are bigger, then the percentage of acrylamide has to be higher as the pore size determines the migration rate, degree of separation of subunits within a macromolecule such as a protein or DNA.
