A student wishes to clone a sequence of DNA of -200kb.Which vector would be appropriate a plasmid a typical bacteriophage a BAC a plant virus a large polypeptide Give the major advantage of using artificial chromosomes such as YACs end BACs for cloning genes. Yeast artificial chromosomes contain which of the following elements? centromeres only telomeres only origin of replication only centromeres and telomeres only centromeres, telomeres, and an origin of replication
Solution
The ideal vector would be a bacterial artificial Chromosome as it would be able to hold upto 350 kb DNA in it while as a plasmid and Bacteriophage would only be able to accomadate less than 50 kb DNA in it. While as Yeast would be able to have a lot more in it.
The major advantage of using BACs and YACs are firstly they have been artificially created by us so they have a particular origin of replication a particular antibiotic that it will be resistant too for us to identify it and the particular endonuclease sites and ligation sites. Along with that they can clone a higher kb of DNA with an easy screening process.
Yeast Artifical Chromosomes have centromeres, telomeres and an origin of replication.
