Plate count method is based on an assumption that each viable organism can produce a bacterial colony. The sample with the bacteria is suspended in the liquid diluent, from this diluent, a known volume is placed in a petri dish.
The cooled sterile agar medium is now poured in the petri dish, mixed and incubated. After incubation, the number of colonies are counted. It is the more frequently used technique to determine the bacterial count.
