  Solution
The SEKDEL sequence of signal peptide leads to retention of the protein in the endoplasmic reticulum of the cell. Hence the protein with GFP and SEKDEL will be seen on endoplasmic reticulum. Since there is no signal peptide in the first protein the protein will be located in the cytoplasm of the cell.
The vertical line can be divided as N terminal signal peptides - gene sequence - C terminal signal peptides. Where the N terminal signals for post translational modification and C terminal signals provides information of protein targeting.
