The improvements to basic cloning vectors like in constructing recombinant plasmids were done in recent years mainly using pBR322 because it has two meticulous antibiotic resistant genes such as the ampR gene to code for recombinant ampicillin resistance protein, and the tetA gene to code for tetracycline resistance protein. It has improved with a “number of suitable restriction sites” and it has prominent replication elements suitable for cloning. Shuttle vectors are constructed even to promote ligation easily at the “blunt ends” using pBR322 therefore, this plasmid is widely improved to use in constructing intraspecies shuttle or binary vectors to generate recombinant proteins.
