An experiment is designed to study the mechanism of sucrose uptake by plant cells. Cells are immersed in a sucrose solution, and the pH of the solution is monitored. Samples of the cells are taken at intervals, and their sucrose concentration is measured. After a decrease in the pH of the solution to a steady, slightly acidic level, sucrose uptake begins.

Solution

1) As it is reported that when the pH drops the uptake of the sucrose increases. so, that means there is pH dependence for the sucrose uptake. we can also say that some type proton cotransporter molecules are involved in this. therefore, we can say that it is a type of active transport.

2) Suitable part of the plant is taken and carefully placed in a cheesecloth bag. This bag is kept in the sucrose solution with labelled C14. The sucrose solution should also be supplemented with the suitable ions required for the growth/activity of that part say cotyledons if taken would also require K+, Ca2+, Mg2+, Na+ etc. Initial pH of the solution is monitored. Theset up is then incubated for a short interval of 15-20 min. small amount of the sample solution of sucrose is collected at suitable intervals of the time. The pH changes are monitored at regular intervals. Simultaneous changes in the concentration of the sucrose in the solution outside the bag would supplement our studies. Now, after, the completion of uptake or uptil when the ph gets stable. the part is taken and heated with Ethanol solution. Now, the tissue is checked for the accumulation of the sucrose.

2. The same experiment is repeated with the addition of the dilute acid to change the pH of the sucrose solution and sucrose concentration accumulated is checked again.

3. the same experiment is repeated with the ATP inhibitor added in the begining to the sucrose solutions.

The readings of each experiment are taken and correlated.

3) Hypothesis: Sucrose transport is pH dependent and therefore must have involved the proton co-transporters.

4) ATP inhibitors would reduce influx by reduction of H+, the driving force for cotransport, either by eliminating the pH

gradient or depolarizing the membrane potential, or both.
