CATALASE TEST 1 Would a false positive from the reaction bet
CATALASE TEST
1. Would a false positive from the reaction between the inoculating loop and hydrogen peroxide be caused by poor specificity or poor sensitivity of the test system? Explain.
OXIDASE TEST
2.Think about theadvice given in the procedure to test a known oxidase-positive organism along with an unknown. a.) Is this a positive or a negative control? b.) What information is provided by the results?
3. Provide a possible explanation as to why the phenylenediamine reagent reduces cytochrome c (aa3) oxidase and not other oxidases (such as cytochrome d of E. coli shown in Fig. 5.18).
STARCH HYDROLYSIS (AMYLASE TEST)
2. How would you expect both positive and negative results to be affected if you were to add glucose to the medium?
I WOULD REALLY APPRECIATE SOME HELP WITH THESE THREE QUESTIONS REGARDING \"MICROBIOLOGY LAB\". I WILL RATE ANSWERS. THANKS A LOT.
Solution
1) No false positive will be made in case of catalase test because the inoculating loop is made with such items that u does not react with the any media or something hence it will not interfere with the results....
3) yes it wil affect both the positive and negative results because in this test th activity of the amylase enzymes is tested on the presence of the glucose formed after breaking amylose into glucose or maltose.. So on adding the glucose from outside it will lead to the increase in the level of amylase activity in positive test and will also give the test results on the negative test on basis of given glucose...!!!
