Curiously, when the cultured mammalian cells were stained with hematoxylin, there were additional structures visible within the nucleus. What were those structures? Is the number of these structures that you observed expected in a normal cell?

To overcome the limitation of optical microscopy staining method can be useful. Hematoxylin is used for the staining of mamalian cell. there is two type of staining regressive and progressive.in both method it binds with negetively charge DNA and RNA. when it form compllex with cellular DNA and RNA it give blue colour. the identified structure may be made of negetive charged DNA and RNA. so it is the frist assumption that it must contian negetive chrged DNA and RNA. now the negetive molecules which is present in nucleas is mostly chromatinous part. it stain mucin and cytoplsama.it also stain golbet cell of intenstine. so as per my knowlege the structure must contain negetively charged DNA and RNA if the test is postive. and the no. of such structure may be very because of false mutation in normal cells .
