DNA fragments of similiar size will not always resolve on a gel. For example.....fragments of 5,804 bp and 5,643 bp will migrate as a single heavy band. Those are referred to as a doublet and can be recognized because they are brighter and thicker than similarly sized singlets. What oculd be done to resolve the doublet fragments?
Solution
Two options are here,
one is capillary electrophoresis, which is so precise it helps in seperation of two DNA strands, and the other one is
the increasing the % of agaraose - It also will seperate the DNA bands.
