Dark-field microscopy is a very simple and effective technique and is used to view live and unstained biological samples. On the specimen, the hollow cone of light is focused in such a way, that the unrefracted and unreflected rays will not enter the objective. So, light which was actually refracted or reflected by the specimen will form the image. Here the field surrounding the specimen will look dark and object will look illuminated. Due to this, we can see the internal structures present in the cell.
