In the normal haemoglobin, Glutamate is the amino acid which is hydrophilic in nature. However, when the mutation occurs, glutamate is replaced with Valine, which is hydrophobic in nature. Thus, mutated heamoglobin moves away from water in blood making it to sickle. This sickle shape does not pass through the spleen and are degraded. Thus, it leads to a lethal condition called sickle cell anemia.
