What are the sequence elements that are associated with wrap
What are the sequence elements that are associated with wrapping DNA around histones, and what are the major changes to the DNA structure that occur here? Remember both AT and GC-rich sites, and say whether this spacing must be EXACT or approximate, explaining why. What is the dyad site in the nucleosome (what does knowing the dyad site predict)?
Solution
Core particles are connected by stretches of linker DNA, which is about 80 bp long. H1 is located in the region of linker DNA and seals DNA in the nucleosomes. Two “Assembly proteins” seem to be involved in construction of nucleasomes from histone and DNA. The two Assembly proteins are nucleoplasmin and N1. In the nucleosome core the average GC content is significantly higher than the average AT content. G/C are symmetric with respect to the dyad. T/A are symmetric with respect to the dyad. Predicted dyad positions at 40bp and 210 bp relative to the start of the 313 bp long repetitive element.
