1. PCR involves process of heating and cooling called thermo cycling. There are three states, they are 1. Denaturing 2. Annealing and 3. Elongation.

2. In these three stages different temperature is being maintained. The order of temperature is as follows.

1. 94oC for 1 minute; At this temperature DNA is denatured.

2. 55oC for 1 minute: At this temperature anneals primer with single stranded DNA.

3. 72oC for 1 minute: This is optimal temperature to synthesize new DNA strand.

4. 94oC for 1 minute; At this temperature again complementary and template DNA is denatured.

5. 55C for 1 minute: At this temperature again anneals primer with both complementary and template DNA.to synthesize new DNA strand.
